Great thread.
Some advice:
Remember to set yourself apart from the crowd.
Keep in mind that there are thousands of people do exactly what you are doing (trying to get a job), therefore do something that will make you stand out.
Example, do some research on the company you are interested in working for, find out the name of the HR Manager/person responsible for hiring, and go hand in your resume in person to him/her. It helps if you arrive early in the morning, when they first open.
It puts a face to your resume and companies usually will notice that you've taken the initiative to get up early, get dressed, and search for a job, as opposed to sitting at home in your pjs while filling out job applications via the internet, like many others are doing.
